The German iSport driver hit the front at half distance as long-time leader Karun Chandhok was spun out of the race by Kazuki Nakajima.
And he then pulled away to win by 9.469 seconds from Brazilian Xandi Negrao - who claimed his first podium finish - and Britain's Adam Carroll in third.
Lucas di Grassi, who began the race as the championship leader, finished down in 11th and now trails Glock by four points in the standings with six races to go.
Polesitter Chandhok led away from the lights in his Durango entry, followed by DAMS driver Nakajima and the slow-starting Minardi-Piquet car of Xandi Negrao.
Glock made it up to fourth and then passed Negrao around the outside at the complex to wrest the final podium spot.
He set about closing in on the lead duo, which he did to make it a three-way fight by half-distance.
But three soon became one on lap 13 as Nakajima made an over-optimistic lunge at Chandhok into the final corner, spinning him out of the race and destroying his chance of becoming the first Indian to win a GP2 race.
With Nakajima also delayed and then retiring with the resultant damage caused to his front end, Glock assumed a lead he would never lose and duly claimed his third win of 2007 and the fifth of his career.
Behind Negrao, there was a raging battle between Borja Garcia and Adam Carroll for fourth, which became third after the incident at the front and was eventually settled in favour of the British FMS driver who made a successful move into the penultimate turn with seven laps to go.
Their battle was briefly joined by both di Grassi and Giorgio Pantano, but the pair dropped back to an eventual 11th and 13th after both were given drive-through penalties for colliding early on.
That allowed Russian Vitaly Petrov, who kept out of trouble all afternoon, to claim an excellent fifth place for Campos ahead of Bruno Senna's Arden entry.
Senna lost sixth place when he lost his rear end under braking while challenging Petrov, allowing Luca Filippi through and into the final points position.
But just when the Italian, who started 24th on the grid, thought he would be given a just reward for his efforts, Senna shot back by with two laps to go to seal the final point.
Roldan Rodriguez and Ho-Pin Tung were ninth and tenth, benefiting from two separate incidents that eliminated some of their competitors, namely local boy Jason Tahinci and Sebastian Buemi after the pair collided at the final corner mid-race.
Ricardo Risatti completed the top ten on only his second GP2 start for Trident, while his team-mate Kohei Hirate collided with Britain's Mike Conway on the opening lap, sending the British driver out of the race.
