The two big-hitters were always likely to be evenly match and so it proved, with three of the four sets in a lengthy match going to tie-breaks.
But the South African Anderson looked the better of the two players for much of the evening when it counted and, as the clock ticked close to 2am, he got it over the line in a nerve-wracking fourth-set tie-break.
Anderson will play Spain's Pablo Carreno Busta in the semi-finals, with Roger Federer and Rafael Nadal both in the other side of the draw at Flushing Meadows.
A Grand Slam semi-final appearance will be unfamiliar territory for the 31-year-old. His best previous effort at one of the four big events of the calendar was a quarter-final appearance in New York back in 2015.
