Nathan Tyson and Andy Williams scored within eight minutes to knock the stuffing out of the Spireites, with substitute Cameron Stewart putting the gloss on an excellent performance with a third goal in injury-time.
"We have got to do better than that. We missed too many opportunities and then we gave a penalty away," said former Rovers boss Saunders after a third straight loss.
"Dan's [Jones] given it away with a handball, which he shouldn't have done, but we had opportunities in the first half. Dan Gardner's had an open goal, side-footed it wide, and then Jay O'Shea missed an open goal. That's two clear-cut chances we missed.
"We felt quite comfortable at half-time and I felt it was just a matter of time before we got on top and they've come out, hustling and bustling and pressing us, but we failed to take another couple of chances and they go and get a penalty. We didn't even follow in for the penalty, which was disappointing, and then they scored a second which meant we were behind the eight-ball.
"We have let eight goals in in three games now and we seem to be getting punished for everything at the moment. There will be changes for Saturday."
After a tame first half in which Gardner and O'Shea went close for Chesterfield and Cedric Evina for Rovers, the match came to life after half-time as Williams and Tyson both saw shots saved by Tommy Lee, while Felipe Mattioni curled one just wide.
Then Rovers were awarded a penalty as Daniel Jones handled Mattioni's attempted flick and despite Lee saving Tyson's spot-kick, the frontman followed up to ram home the rebound.
Eight minutes later, Williams combined brilliantly with Evina, to start and finish a move which ended with the marksman sweeping the ball in from 10 yards.
Williams nearly had a second, but prodded sub Cameron Stewart's cross wide, and after O'Shea saw an effort palmed clear by Thorsten Stuckmann, Stewart himself then broke clear to finish with aplomb at the death.
Following Rovers' 2-0 defeat to Rochdale at the weekend, Doncaster boss Darren Ferguson admitted he was pleased to see his side hit back in style.
"It was a great way to bounce back. They started well, but after about 10 minutes we dominated and could have had more than three goals," said the Scot.
"It was a good display, another clean sheet and it makes last Saturday's performance even more annoying. It puts us in an even more positive frame of mind.
"Our pressing game worked well, I felt technically the players were good and I am very pleased."
On his goal, Tyson added: "It's an added bonus and about time. It's the first career penalty I've ever missed and my heart was in my mouth when I he saved it, but luckily I followed it in.
"Not scoring goals had been playing on my mind for a few weeks and I needed that goal tonight."
